Osłabiona indukcja odpowiedzi niezfałdowanego  białka  w astrocytach pierwotnych człowieka dorosłego w odpowiedzi na nawracające niskie stężenie glukozy

Cele/hipoteza:  Nawracająca hipoglikemia (RH) jest głównym skutkiem ubocznym intensywnej insulinoterapii u osób z cukrzycą. Zmiany w wykrywaniu hipoglikemii przez mózg przyczyniają się do rozwoju zaburzonych odpowiedzi kontrregulacyjnych i świadomości hipoglikemii. Niewiele wiadomo na temat wewnętrznych zmian w ludzkich astrocytach w odpowiedzi na ostrą i nawracającą ekspozycję na niski poziom glukozy (RLG).

Metody:  Ludzkie pierwotne astrocyty (HPA) wystawiono na zero , jeden, trzy lub cztery ataki niskiego poziomu glukozy (0,1 mmol/l) przez trzy godziny dziennie przez cztery dni, aby naśladować RH. Czwartego dnia pobrano DNA i RNA. Różnicową ekspresję genów i analizy ontologiczne przeprowadzono odpowiednio przy użyciu DESeq2 i GOseq. Metylację DNA oceniano za pomocą platformy Infinium MethylationEPIC BeadChip.

Wyniki:  Wykryto 24 geny o zróżnicowanej ekspresji (DEG) (po korekcie na wielokrotne porównania). Jeden atak niskiej ekspozycji na glukozę miał największy wpływ na ekspresję genów. Analizy szlaków wykazały, że geny związane ze stresem retikulum endoplazmatycznego (ER), takie jak  HSPA5 ,  XBP1 i  MANF , zaangażowane w odpowiedź na niesfałdowane białka (UPR), były znacząco zwiększone po ekspozycji na niski poziom glukozy (LG), która uległa zmniejszeniu po RLG . Istniała niewielka korelacja między pozycjami zróżnicowaną metylacją a zmianami w ekspresji genów, jednak liczba ataków ekspozycji na LG dała wyraźne sygnatury metylacji.

Wnioski/interpretacja:  Dane te sugerują, że ekspozycja ludzkich astrocytów na przemijające LG wyzwala aktywację genów zaangażowanych w UPR związane ze stresem retikulum endoplazmatycznego (ER). Po RLG aktywacja genów związanych z UPR została zmniejszona, co sugeruje osłabiony stres ER. Może to być konsekwencją udanej adaptacji metabolicznej , jak wcześniej zgłoszono , która lepiej zachowuje wewnątrzkomórkowe poziomy energii i zmniejszoną konieczność UPR.

Doksorubicyna/ białko wiążące  nukleofosminę – sprzężone nanocząstki zwiększają aktywność przeciwbiałaczkową w komórkach ostrej białaczki limfoblastycznej  in vitro  i  in vivo

Ostra białaczka limfoblastyczna (ALL) to agresywny nowotwór złośliwy. Dorośli z ALL mają ponad 50% wskaźników nawrotów. Wcześniej potwierdziliśmy, że nadekspresja nukleofosminy (NPM) jest zaangażowana w rozwój oporności wielolekowej (MDR) podczas ALL ; aw naszej grupie opracowano syntetycznie modyfikowane rekombinowane białko wiążące NPM (NPMBP); NPMBP i doksorubicyna (DOX) mogą być sprzężone w systemie dostarczania leków opartym na nanocząstkach o nazwie DOX-PMs-NPMBP, aby przeciwdziałać MDR podczas ALL. Tutaj oceniliśmy potencjał przeciwbiałaczkowy DOX-PMs-NPMBP w opornych komórkach ALL. Badanie to pokazuje, że DOX-PMs-NPMBP znacząco zwiększa chemowrażliwość na DOX w komórkach ALL.

Pomimo różnych stężeń, zarówno oporne, jak i pierwotne komórki ALL od pacjentów z nawrotem były wrażliwe na DOX-PMs-NPMBP . W szczególności, wartości połowy maksymalnego stężenia hamującego (IC50) DOX-PMs-NPMBP były od 1,6 do 7,Zero razy niższe niż te dla DOX odpowiednio w liniach komórkowych i pierwotnych komórkach ALL; a stosunek komórek apoptotycznych był ponad 2-krotnie wyższy w DOX-PMs-NPMBP niż w DOX.

Pod względem mechanicznym indukcja apoptozy wywołana przez p53 i zatrzymanie cyklu komórkowego odegrały zasadniczą rolę w efektach przeciwbiałaczkowych indukowanych przez DOX-PMs-NPMBP. Ponadto DOX-PMs-NPMBP znacząco hamował wzrost guza i przedłużył przeżycie myszy w modelach heteroprzeszczepu ALL; i nie systemowywystąpienie toksyczności zaobserwowano po leczeniu podczas obserwacji. Podsumowując, dane te wskazują, że DOX-PMs-NPMBP może znacząco wywierać hamowanie wzrostu i indukcję apoptozy oraz znacząco poprawiać aktywność przeciwbiałaczkową DOX w opornych komórkach ALL. Ten nowy system dostarczania leków może być cenny do opracowania jako nowa strategia terapeutyczna przeciwko wielolekoopornej ALL.
